Chemistry

Naphazoline Hydrochloride: C~14~H~14~N~2~ HCl. Mw: 246.74. (1) 1H-Imidazole, 4,5-dihydro-2-(1-naphthalenylmethyl)-, monohydrochloride; (2) 2-(1-Naphthylmethyl)-2-imidazoline monohydrochloride. CAS-550-99-2; CAS-835-31-4 (naphazoline).

Pharmacologic Category

EENT Preparations; Vasoconstrictors; α~1~-Adrenergic Agonist; Imidazoline Derivative. Ophthalmic Agent; Vasoconstrictor. (ATC-Code: R01AA08; R01AB02; S01GA01).

Mechanism of action

Stimulates α-adrenergic receptors in arterioles of conjunctiva and nasal mucosa to produce vasoconstriction.

Therapeutic use

Topical ocular vasoconstrictor. Temporary relief of nasal congestion associated with common cold, upper respiratory allergies or sinusitis. Relief of redness of eye due to minor irritation.

Pregnancy and lactiation implications

Naphazoline hydrochloride should be used during pregnancy only when clearly needed. Animal reproduction studies not conducted. Excretion in breast milk unknown (use caution).

Unlabeled use

Contraindications

Hypersensitivity to naphazoline or any component of the formulation. Angle-closure glaucoma.

Warnings and precautions

Rebound nasal congestion might occur with extended use. Use with caution in chronic asthma, hypertension, heart failure, coronary artery disease, cerebral arteriosclerosis, diabetes mellitus, hyperthyroidism, local infection or injury, and in benign prostatic hyperplasia. Avoid concurrent use with MAOIs (may cause hypertensive crisis). Not recommended for use in children <6 years of age. Excessive dosage may cause CNS depression in children, coma and marked reduction in body temperature. Ophthalmic products may contain benzalkonium chloride.
