Pharmacologic Category

Mechanism of action
Inhibits steroid synthesis by blocking the enzymatic conversion of cholesterol to δ-5-pregnenolone.
Therapeutic use
Suppression of adrenal function in Cushing’s syndrome.
Pregnancy and lactiation implications
May cause fetal harm. Suspected of causing virilization. Administration is not recommended during lactation.
Unlabeled use
Treatment of prostate cancer (androgen synthesis inhibitor).
Contraindications
Hypersensitivity to aminoglutethimide, glutethimide, or any component of the formulation.
Warnings and precautions
Adrenal hypofunction may occur (particularly under conditions of stress, including acute illness, surgery, and/or trauma). May cause drowsiness. Hematologic abnormalities (neutropenia, leukopenia, pancytopenia and agranulocytosis) have been reported. May cause persistent or orthostatic hypotension (due to suppression of aldosterone production). Hypothyroidism might occur.
