Pharmacologic Category

Antineoplastic Agents; Endocrine Therapy. Aromatase Inhibitor. (ATC-Code: L02BG03).

Mechanism of action

Inhibits aromatase, thus preventing the conversion of androstenedione to estrone, and of testosterone to estradiol.

Therapeutic use

Treatment of locally-advanced or metastatic breast cancer in postmenopausal women. Treatment of advanced breast cancer in postmenopausal women with disease progression following tamoxifen therapy. Adjuvant treatment of early estrogen receptor-positive breast cancer in postmenopausal women.

Pregnancy and lactiation implications

Fetotoxicity was observed in animal studies. Exclude pregnancy before initiating. Use with caution during lactation.

Contraindications

Hypersensitivity to anastrozole or any component of the formulation. Pregnancy.

Warnings and precautions

Hazardous agent. May cause decreases in bone mineral density. Use with caution in hyperlipidemias (total cholesterol and LDL-cholesterol increase). Exclude pregnancy before initiating.
