Baclofen: C~10~H~12~ClNO~2~. Mw: 213.66. (1) Butanoic acid, 4-amino-3-(4-chlorophenyl)-; (2) β-(Aminomethyl)-p-chlorohydrocinnamic acid. CAS-1134-47-0 (1971).

Pharmacologic Category

GABA-derivative Skeletal Muscle Relaxants. (ATC-Code: M03BX01).

Mechanism of action

Inhibits the transmission of mono/polysynaptic reflexes at the spinal cord level, possibly by hyperpolarization of primary afferent fiber terminals.

Therapeutic use

Treatment of spasticity associated with spinal cord lesions.

Pregnancy and lactiation implications

Crosses placenta. Enters breast milk (small amounts). Compatible with breast-feeding.

Unlabeled use

Bladder spasticity, cerebral palsy, intractable hiccups, intractable pain relief, trigeminal neuralgia, Huntington’s chorea.

Contraindications

Hypersensitivity to baclofen or any component of the formulation.

Warnings and precautions

Withdrawal reactions may occur with abrupt discontinuation, particularly severe with intrathecal use (hyperpyrexia, obtundation, rebound/exaggerated spasticity, muscle rigidity, and rhabdomyolysis), leading to organ failure and fatalities. Risk of CNS depression. Caution in history of seizure disorder. Caution in renal disorders.
