Danaparoid Sodium: Danaparoid is a low molecular weight heparinoid, a mixture of the sodium salts of heparan sulfate (approx. 84%), dermatan sulfate (approx. 12%), and chondroitin sulfate (approx. 4%). It is derived from porcine intestinal mucosa. Heparan Sulfate: Heparitin, sulfate, sodium salt. CAS-57459-72-0. Dermatan Sulfate: Dermatan, 4-(hydrogen sulfate), sodium salt. CAS-54328-33-5. Chondroitin 4-sulfate: Chondroitin, 4-(hydrogen sulfate), sodium salt. CAS-39455-18-0. Chondroitin 6-sulfate: Chondroitin, 6-(hydrogen sulfate), sodium salt. CAS-12678-07-8 (1992).

Pharmacologic Category

Antithrombotic Agents; Anticoagulants; Heparins. (ATC-Code: B01AB09).

Mechanism of action

Prevents fibrin formation in coagulation pathway via thrombin generation inhibition by anti-Xa and anti-IIa effects.

Therapeutic use

Prevention of postoperative deep vein thrombosis following elective hip replacement surgery.

Pregnancy and lactiation implications

Compatible with lactation.

Unlabeled use

Systemic anticoagulation for heparin-induced thrombocytopenia.

Contraindications

Hypersensitivity to danaparoid. Thrombocytopenia. Hypersensitivity to pork products or sulfites. Hemorrhagic diathesis. Not for I.M. or I.V. use.

Warnings and precautions

Patients with certain risk factors may be at risk of bleeding. May cause hyperkalemia by affecting aldosterone. Caution in hypersensitivity to methylparaben or propylparaben, history of heparin-induced thrombocytopenia or severe renal failure. Contains sodium sulfite, which may cause allergic-type reactions (frequently seen in asthmatics). Not convertible to other low molecular weight heparins. Risk of spinal or epidural hematoma in recent or anticipated neuraxial anesthesia.
