Chemistry

Mecamylamine Hydrochloride: C~11~H~21~N HCl. Mw: 203.75. (1) Bicyclo[2.2.1]heptan-2-amine, N,2,3,3-tetramethyl-, hydrochloride; (2) N,2,3,3-Tetramethyl-2-norbornanamine hydrochloride. CAS-826-39-1; CAS-60-40-2 (mecamylamine).

Pharmacologic Category

Hypotensive Agents, Miscellaneous; Ganglionic Blocking Agent. (ATC-Code: C02BB01).

Mechanism of action

Inhibits acetylcholine at autonomic ganglia, causing decrease in blood pressure. Also blocks central nicotinic cholinergic receptors, which inhibits effects of nicotine and may suppress desire to smoke.

Therapeutic use

Treatment of moderately severe to severe hypertension and in uncomplicated malignant hypertension.

Pregnancy and lactiation implications

Unlabeled use

Tourette’s syndrome.

Contraindications

Coronary insufficiency, pyloric stenosis, glaucoma, uremia, recent MI.

Warnings and precautions

Use with caution in previous CNS abnormalities, in prostatic hyperplasia, bladder obstruction, or urethral stricture (might cause urinary retention), and in renal impairment. Do not discontinue abruptly.
