Chemistry

Meclizine Hydrochloride: C~25~H~27~ClN~2~ 2HCl H~2~O. Mw: 481.89. (1) Piperazine, 1-[(4-chlorophenyl)phenylmethyl]-4-[(3-methylphenyl)methyl]-, dihydrochloride, monohydrate; (2) 1-(p-Chloro-a-phenylbenzyl)-4-(m-methylbenzyl)piperazine dihydrochloride monohydrate. CAS-31884-77-2; CAS-1104-22-9 (anhydrous); CAS-569-65-3 (meclizine).

Pharmacologic Category

Antiemetics; Antihistamines; First Generation Histamine H~1~ Antagonist. (ATC-Code: R06AE05).

Mechanism of action

Has central anticholinergic action by blocking chemoreceptor trigger zone. Decreases excitability of middle ear labyrinth and blocks conduction in middle ear vestibular-cerebellar pathways.

Therapeutic use

Prevention and treatment of symptoms of motion sickness. Management of vertigo with diseases affecting vestibular system.

Contraindications

Hypersensitivity to meclizine or any component of the formulation.

Warnings and precautions

Might cause CNS depression. Use with caution in asthma. Use with caution in narrow-angle glaucoma (might be exacerbated by cholinergic blockade). Use with caution in prostatic hyperplasia and/or urinary stricture. Use with caution in pyloric or duodenal obstruction. Use with caution in the elderly. Effects might be potentiated when used with other sedative drugs or ethanol. In case of lack of response it is advised to discontinue use. Some formulations contain tartrazine dye (may cause allergic reactions).
