Chemistry

Metaxalone: C~12~H~15~NO~3~. Mw: 221.25. (1) 2-Oxazolidinone, 5-[(3,5-dimethylphenoxy)methyl]-; (2) 5-[(3,5-Xylyloxy)methyl]-2-oxazolidinone. CAS-1665-48-1 (1962).

Pharmacologic Category

Centrally Acting Skeletal Muscle Relaxants. (ATC-Code: M03BB).

Mechanism of action

Efficacy appears to result from disruption of spasm-pain-spasm cycle, probably by general CNS-depressant effect. Does not have direct effect on skeletal muscle.

Therapeutic use

Relief of discomfort associated with acute, painful musculoskeletal conditions.

Pregnancy and lactiation implications

Do not use in women who are or may become pregnant unless possible benefits outweigh potential risks. Use not recommended during lactation.

Unlabeled use

Contraindications

Hypersensitivity to metaxalone or any component of the formulation. Impaired hepatic or renal function, history of drug-induced hemolytic anemias or other anemias.

Warnings and precautions

Might cause CNS depression. Use with caution in hepatic or renal impairment. Effects might be potentiated when used with other sedative drugs or ethanol. Use with caution in the elderly (might be more sensitive to CNS effects). Increase in bioavailability and half-life observed in female patients.
