Pharmacologic Category

Antiglaucoma Agents; Carbonic Anhydrase Inhibitors. Diuretic. (ATC-Code: S01EC05).

Mechanism of action

Noncompetitive inhibition of enzyme carbonic anhydrase. When enzyme is inhibited, there is increase in urine volume and change to alkaline pH with subsequent decrease in excretion of titratable acid and ammonia.

Therapeutic use

Adjunctive treatment of open-angle or secondary glaucoma. Short-term therapy of narrow-angle glaucoma when delay of surgery is desired.

Pregnancy and lactiation implications

Use with caution during pregnancy or lactation.

Unlabeled use

Contraindications

Hypersensitivity to methazolamide or any component of the formulation. Marked kidney or liver dysfunction. Severe pulmonary obstruction.

Warnings and precautions

Might produce CNS effects. Use in sulfonylurea allergy contraindicated. Use with caution in prediabetes or diabetes mellitus (possible change in glucose control), in pulmonary obstruction, emphysema, or advanced pulmonary disease (might precipitate or aggravate acidosis), and in the elderly. Avoid use in marked hepatic impairment, including cirrhosis (risk of developing hepatic encephalopathy), and in marked renal impairment.
