Chemistry

Pancrelipase: A concentrate of pancreatic enzymes standardized for lipase content. (1) Lipase, triacylglycerol; (2) Lipase of pancreas. CAS-53608-75-6 (1963).

Pharmacologic Category

Gastrointestinal Drugs; Digestants. Enzyme. (ATC-Code: H04).

Mechanism of action

Contains combination of lipase, amylase, and protease. Products formulated to dissolve in more basic pH of duodenum so they may act locally to break down fats, protein, and starch.

Therapeutic use

Replacement therapy in symptomatic treatment of malabsorption syndrome caused by pancreatic insufficiency.

Pregnancy and lactiation implications

Use caution during pregnancy or lactation.

Unlabeled use

Treatment of occluded feeding tubes.

Contraindications

Hypersensitivity to pork protein or any component of the formulation. Acute pancreatitis or acute exacerbations of chronic pancreatic disease.

Warnings and precautions

Fibrotic strictures in colon (some requiring surgery) reported with high doses; use caution, especially in children with cystic fibrosis. Pancrelipase inactivated by acids. Avoid inhalation of powder (may cause nasal and respiratory tract irritation).
