Tazarotene: C~21~H~21~NO~2~S. Mw: 351.46. (1) 3-Pyridinecarboxylic acid, 6-[(3,4-dihydro-4,4-dimethyl-2H-1-benzothiopyran-6-yl)ethynyl]-,ethylester; (2) Ethyl 6-[(4,4-dimethylthiochroman-6-yl)ethynyl]nicotinate. CAS-118292-40-3 (1994).

Pharmacologic Category

Skin and Mucous Membrane Agents, Miscellaneous. Keratolytic Agent. Topical Skin Product (Acne). (ATC-Code: D05AX05).

Mechanism of action

Synthetic, acetylenic retinoid which modulates differentiation and proliferation of epithelial tissue and exerts some degree of anti-inflammatory and immunological activity.

Therapeutic use

Topical treatment of facial acne vulgaris. Topical treatment of stable plaque psoriasis. Mitigation (palliation) of facial skin wrinkling, facial mottled hyper-/hypopigmentation, and benign facial lentigines.

Pregnancy and lactiation implications

May cause fetal harm if administered to a pregnant woman. Excretion in breast milk unknown (use with caution in nursing women).

Unlabeled use

Contraindications

Hypersensitivity to tazarotene, other retinoids or vitamin A derivatives (isotretinoin, tretinoin, etretinate), or any component of the formulation. Pregnancy.

Warnings and precautions

May cause photosensitivity. Risk may be increased by concurrent therapy with known photosensitizers (thiazides, tetracyclines, fluoroquinolones, phenothiazines, sulfonamides). Treatment can increase skin sensitivity to weather extremes of wind or cold. Also, concomitant topical medications (e.g. medicated or abrasive soaps, cleansers, or cosmetics with a strong drying effect) should be used with caution due to increased skin irritation. Potential teratogenicity. For external use only. Not for use on eczematous, broken, or sunburned skin. Not for treatment of lentigo maligna.
