Chemistry

Vincamine: C~21~H~26~N~2~O~3~. Mw: 354.44. Alkaloid obtained from Vinca minor L. CAS-1617-90-9.

Pharmacologic Category

Vasodilating Agents, Miscellaneous. Peripheral vasodilator. (ATC-Code: C04AX07).

Mechanism of action

Induces relaxation of vascular and visceral musculature responses to angiotensin II. There is no specific antagonism between vincamine and angiotensin II, but its mechanism of action probably lies in its capacity to modify flow of calcium across the cell membrane.

Therapeutic use

Treatment of a variety of conditions related with insufficient blood flow to brain, including vertigo and Meniere’s syndrome, difficulty in sleeping, mood changes, depression, hearing problems, high blood pressure and lack of blood flow to eyes. Also used to improve memory defects and ability to concentrate.

Pregnancy and lactiation implications

Avoid use in pregnant or breast-feeding women.

Contraindications

Cerebral neo-formation with intracranial hypertension.

Warnings and precautions

Rarely causes GI distress, which disappears when use is stopped. Use caution in history of allergic phenomena during treatment with other vinca alkaloids, in patients with hepatic disease, and in those with nonorganic psychoses. Monitor patients with seizure disorder closely.
