Pharmacologic Category
Second Generation Antihistamines; Histamine H~1~ Antagonist. (ATC-Code: R06AX27).
Mechanism of action
An active metabolite of loratadine. Long-acting tricyclic antihistamine with selective peripheral histamine H~1~-receptor antagonistic activity and additional anti-inflammatory properties.
Therapeutic use
Desloratadine, alone or in fixed combination with pseudoephedrine sulfate, is used to provide symptomatic relief of nasal and non-nasal symptoms of seasonal allergic rhinitis (e.g. hay fever), and perennial (nonseasonal) allergic rhinitis. Has also been used in patients with seasonal allergic rhinitis who have concomitant mild to moderate asthma. Also used for the treatment of chronic idiopathic urticaria and relief of symptoms of chronic idiopathic urticaria (e.g. pruritus, hives) in pediatric patients. Desloratadine also suppresses the release of histamine from human mast cells.
Pregnancy and lactiation implications
There are no adequate, well-controlled studies in pregnant women. Use during pregnancy only if clearly needed. Not recommended during lactation.

Contraindications
Hypersensitivity to desloratadine, loratadine, or any component of the formulation.
Warnings and precautions
Some products may contain phenylalanine. Caution in children <2 years of age and in the elderly. Avoid use of the fixed-combination preparation containing desloratadine and pseudoephedrine in hepatic impairment (decreased clearance and increased elimination half-life). Caution in renal impairment and in slow metabolizers of the drug.
