Ciclopirox Olamine: C~12~H~17~NO~2~ C~2~H~7~NO. Mw: 268.35. (1) 2(1H)-Pyridinone, 6-cyclohexyl-1-hydroxy-4-methyl-, compound with 2-aminoethanol (1:1); (2) 6-Cyclohexyl-1-hydroxy-4-methyl-2(1H)-pyridone compound with 2-aminoethanol (1:1). CAS-41621-49-2 (1975).
Pharmacologic Category
Skin and Mucous Membrane Agents; Antifungals; Hydroxypyridones. Topical. (ATC-Code: D01AE14; G01AX12).
Mechanism of action
Causes intracellular depletion of essential substrates and/or ions by inhibiting transmembrane transport of these substances into cells, decreasing protein, RNA, and DNA synthesis. May chelate polyvalent cations (i.e. iron, aluminum) which may result in inhibition of metal-dependent enzymes for the degradation of peroxides within the fungal cell. Active in vitro against Trichophyton equinum, T. mentagrophytes, T. rubrum, T. schoenleinii, T. tonsurans, T. verrucosum, T. violaceum, E. floccosum, Microsporum audouinii, M. canis, C. albicans, and M. furfur (P. orbiculare).
Therapeutic use
Tinea pedis (athlete’s foot), tinea cruris (jock itch), tinea corporis (ringworm), cutaneous candidiasis, and tinea versicolor (pityriasis), seborrheic dermatitis of the scalp, onychomycosis.
Pregnancy and lactiation implications
Teratogenic effects not observed in animal studies. No adequate studies in pregnant women (use during pregnancy only if clearly needed). Excretion in breast milk unknown.

Contraindications
Hypersensitivity to ciclopirox or any component of the formulation. Avoid occlusive wrappings or dressings.
Warnings and precautions
Discontinue if sensitivity or irritation occurs. Topical cream and suspension not approved for use in children <10 years of age. Nail lacquer (solution) not approved for use in children <12 years of age. Shampoo and gel not approved for use in children <16 years of age. Nail lacquer only for topical use.
