Pharmacologic Category

Antibacterials; Second Generation Cephalosporins; Carbacephem Antibiotic. (ATC-Code: J01DC08).

Mechanism of action

Inhibits bacterial cell wall synthesis by binding to one or more penicillin-binding proteins. Inhibits final transpeptidation step of peptidoglycan synthesis in bacterial cell walls, thus inhibiting cell wall biosynthesis.

Therapeutic use

Treatment of mild-to-moderate community-acquired infections of respiratory tract, skin and skin structure, and urinary tract caused by susceptible S. pneumoniae, H. influenzae, M. catarrhalis, S. aureus, S. pyogenes, and E. coli.

Pregnancy and lactiation implications

Unknown whether distributed into milk (use with caution).

Contraindications

Hypersensitivity to loracarbef, any component of the formulation, or cephalosporins.

Warnings and precautions

Possible emergence and overgrowth of nonsusceptible organism. Hypersensitivity reactions such as anaphylaxis, serum sickness-like reactions, Stevens-Johnson syndrome, and toxic epidermal necrolysis reported. Partial cross-allergenicity among β-lactam antibiotics, including penicillins, cephalosporins, and cephamycins. Use with caution in hypersensitivity to penicillins (contraindicated in those hypersensitive to cephalosporins). Use with caution in history of GI disease, particularly colitis. Use with caution in renal impairment.
