Chemistry

Loratadine: C~22~H~23~ClN~2~O~2~. Mw: 382.88. (1) 1-Piperidinecarboxylic acid, 4-(8-chloro-5,6-dihydro-11H-benzo[5,6]cyclohepta[1,2-b]pyridin-11-ylidene)-, ethyl ester; (2) Ethyl 4-(8-chloro-5,6-dihydro-11H-benzo[5,6]cyclohepta[1,2-b]pyridin-11-ylidene)-1-piperidinecarboxylate. CAS-79794-75-5 (1986).

Pharmacologic Category

Second Generation Antihistamines; Histamine H~1~ Antagonist. (ATC-Code: R06AX13).

Mechanism of action

Long-acting tricyclic antihistamine with selective peripheral histamine H~1~-receptor antagonistic properties.

Therapeutic use

Relief of nasal and non-nasal symptoms of seasonal allergic rhinitis. Treatment of chronic idiopathic urticaria.

Pregnancy and lactiation implications

Not found teratogenic in animal studies. Use during pregnancy only if clearly needed.

Contraindications

Hypersensitivity to loratadine or any component of the formulation.

Warnings and precautions

Use with caution in hepatic or renal impairment. Some products may contain phenylalanine.
